While 1 playthrough on Hard difficulty is certainly possible, for the sake of making the mission specific trophies easier and more manageable, I recommend 2 playthroughs - 1 on Normal and 1 on Hard - mainly because on Hard difficulty you get fewer Command Points - 4 instead of the maximum of 6 - as well as the introduction of a timer which forces each turn to be completed within 2 minutes.
Besides being able to manually save during each turn, except when playing on Hard difficulty, there is also a very convenient undo mechanic. To undo an action simply press . This is incredibly helpful as it allows you to perform and undo an action until the desired outcome is achieved. Please note that you cannot make use of the undo mechanic after ending a turn. Also, I strongly suggest playing through the tutorial because it gives a much needed explanation on the various game mechanics.
Step 1: Complete the main campaign "Sin of Damnation" on Normal difficulty
Play through the campaign whilst simultaneously going for as many of the mission specific trophies as possible. Some of the mission specific requirements can be a little tricky. However, as each and every mission can be replayed through mission select after being completed once, you can either decide to replay a mission until unlocking a certain mission specific trophy or you can storm through the campaign and return later via mission select.
As for the various miscellaneous trophies, most will unlock through natural gameplay. Just as with the mission specific trophies, you can either decide to get them out of the way as soon as possible or you can go for any remaining via mission select.
Step 2: Complete the main campaign "Sin of Damnation" on Hard difficulty
On this playthrough you should focus solely on completing the main objectives. You now only have a maximum of 4 CPs instead of 6; there is a timer introduced which forces you to complete each turn within 2 minutes; manual saving in a mission is prohibited although the undo mechanic is still available.
Rely heavily on Overwatch and be sure to only spend any remaining CPs when it is safe to do so. On missions where the Librarian is available, make good use of the Force Barrier in order to prevent the Genestealers from flanking. Also, using the Heavy Flamer equipped Terminator as lead unit is tactically sound, especially so when speed is of the essence and you quickly need to advance with minimal delay.
