Road Map
There isn't much to write for a road map here. This game's trophies can mostly be done in exhibition games. You don't need to really explore the other features of the game, other than online. It's really all about setting up games that can maximize your trophies and get them all as quickly as possible. The trophy descriptions are specific about the settings, so you should follow them exactly. If it says "5 minute quarters", then don't do 6, 7, or 15 minute quarters. Set it at 5.
There are a few things that you can do to make things much easier for you. Yes, the difficulty should be set to All Pro. You'll need it there for most of the trophies anyway. There are a few where you can set it to Rookie, but for the most part it should always stay the same. Even though this is the second highest difficulty in the game, the trophies are still pretty easy because you can adjust the AI sliders to give yourself as big of an advantage as you want. You can also edit the opposing player's attributes to make them less skilled and more likely to do what you want them to do so you can get your trophies. You can also use two controllers. Yes, the trophies say "No Co-op". But that just means two people playing on the same team. You can have two controllers set to different teams.
Here is a game that you should set up to maximize your time and get the trophies in the most efficient way possible.
Arizona Cardinals (you) vs. San Diego Chargers in Lambeau Field:
Set the weather to Heavy Snow. Player one is the Cardinals and player two is the Chargers. Have the Chargers take an early lead until the last 30 seconds of the game. Between then, mess around getting trophies.
Trophies you can get: Sprint Two-Minute Drill, Super Bowl MVP-Like, Frozen Tundra, Go Back to Basketball, Nothing Without the Dark Visor, Fight for the Fumble, Look What I Found!, Larry Stickyfingers. Those are the team and situation specific ones, but you can also get Nano, Pocket Monster, Well Played, Really and Wounded Duck in the game since those only take one play.
Trophy Clean Up
Most of the rest of trophies will come one at a time. There aren't many times you can get multiple trophies in a game (with the exception of playing against the New England Patriots). Play the remaining games against the Indianapolis Colts, Arizona Cardinals, and Minnesota Vikings.
After all of the game trophies are completed, complete the two Creation trophies. Those will take about five minutes to do. After the Creation trophies are finished, head over to the Co-Op partner thread to find yourself a partner and complete those three trophies. All you have to do is complete one game, but you have to complete a lateral and a touchdown pass.
